The fourth term will raise x^2 to the 2 and (1/4x)^4.  That will give x^4 in both the numerator and denominator.
The leading term is 10 (1-5-10-10-5-1 in expansion to 6th power)
10*(3x^2)^2*(1/4x)^4
=10*9x^4*(1/256x^4)=90/256, or 45/128.
