No it does not. Just take any two coordinates and try them both ways, the slope will be the same always.
For example (5, -1) and (3, 6)
m=(y - ysub1)/(x - xsub1)
(-1-6)/(5-3)=(-7)/2 or -(7/2)
(6-(-1))/(3-5)=7/(-2) or -(7/2)
Here it doesn't matter where the negative is, either in the numerator or the denominator, because both ways its show a negative slope
