I think the answer to your question is 'Yes' (as long as the rise and the run are both positive.
In other words, slope = rise/run
When the slope is 3/1, then you move up 3 units (in the positive direction) and to the right 1 unit (in the positive direction).
If the slope = -3/1, then the negative sign can go before the rise or it can go before the run.
In other words, slope = (-3)/1 means you go down 3 units (because the rise is negative), and you go to the right 1 units (because the run is positive).
If slope = 3/(-1), then you go up 3 units (because the rise is positive), and you go to the left 1 unit (because the run is negative).
