Assuming the 2x4s are 6 feet long (standard - could also be 8'), and assuming you mean 50' across with 12" on center between the boards, you'll need 51 boards.
Start at one side and place boards 12" on center from each other.
HOWEVER, if you mean a 12" space inbetween and not on-center, then you need 38 boards. You'll start at one end with a 4" board + 12" + 4" + 12", etc.
If this is a real-life situation, realize that a 2x4 is not actually 2"x4" any more. After drying, it's 1.5" x 3.5". Also, you should space on-center -- 12" from the middle of the board to the next middle of the board, not 12" between.
