$5 million dollars in 20 dollar bills would be 250000 bills.

From Wikipedia, each bill is 2.6" x 6.1" and 0.004" thick and weighs 1 gram.
So, the bills would weigh 250 kg, a bit much for a briefcase.

Also, a briefcase is approximately 18x12x4 in dimensions which gives a total volume of 864 cubic inches. The volume of the bills would be 6.14x2.61x.0043x250000 which equals 17227 cubic inches. So it's highly unlikely that the bills will fit in any normal briefcase.
