Directions: Using figure 7.1, measure the distance from the loading well to each band on your gel and set up a table with your results on the worksheet titled Assignment 7.3. Next, use algebra to determine the log of unknown fragments in proportion to a logarithmic scale. Then, using the graph on the worksheet titled Assignment 7.3, plot the points for each of the standards and draw a straight line that connects as many of these as possible. Use the X-axis to plot migration distance and the Y-axis to plot the log of the size or molecular weight. Now you can take the distance of migration for any unknown sample and find that point on the X-axis; then go across to the regression line; and from that point, go down to the Y-axis. This point will represent the log of the size of your unknown band.
