It is possible. One example would be to have points at
A = (3, 3)
B = (4, 2)
C = (3, 1)
D = (2, 2)

Quadrilateral ABCD is a square with side lengths sqrt(2), which can be found using either the pythagorean theorem or the distance formula.

The area of ABCD is (sqrt(2))^2 = 2 square units. Point E = (3,2) is the only interior point such that both coordinates are integers.

Any translation, reflection and/or rotation of ABCD will also be a valid example as well.
